| 摘 要： | 目的:观察大肠癌细胞突变的线粒体DNA(mtDNA)转染小鼠胚胎成纤维细胞NIH3T3后,mtDNA与转染细胞核内基因组的整合情况.方法:将携带大肠癌细胞突变mtDNA的重组质粒pcDNA3.1( )-Sw480 mtDNA、pcDNA3.1( )-NHWBC mtDNA及空质粒pcDNA3.1( )通过脂质体法转染NIH3T3细胞.同时用PCR法制备3条相应的绿色荧光蛋白标记的mtDNA探针.分离转染的NIH3T3细胞核内染色体,用荧光原位杂交(FISH)法检测mtDNA探针与核内基因组的整合情况.结果与结论:瞬时转染的NIH3T3细胞核基因组上存在mtDNA的同源序列.外源肿瘤细胞mtDNA与NIH3T3细胞核内基因组发生整合. |

| Abstract: | Aim: To observe the integration of mtDNA of colorectal carainoma cells and transfected cells nuclear genome.Methods:Plasmid pcDNA3.1( )-SW480 mtDNA, pcDNA3.1( )-NHWBC mtDNA, and pcDNA3.1( ) were transfected into NIH3T3 cell line using lipofectamine. Three kinds of non-radioactive DIG labelled mtDNA probes were prepared by PCR, and the chromosomes of transfected NIH3T3 cells were analyzed by fluorescence in situ hybridization. Results and Conclusion: The mutated mtDNA of human colorectal carcinoma cells were integrated into the nuclear genome of transfected NIH3T3 cell. |
