| 摘 要： | 本文报告了一个从携带者血清中大量提取HBsAg的方法。首先,用5.5%聚乙二醇6000使Dane颗粒和主要的脂蛋白沉淀;第二步用11%聚乙二醇把HBsAg沉淀出来;第三步用溴化钾作漂浮密度梯度区带离心,和第四步用蔗糖密度梯度速率区带纯化所得的HBsAg光密度单位剩余为0.32%,也就是去除了99.68%杂蛋白,回收率为73.4%。

| Abstract: | This paper report, a combined method of extracting large amount from HBsAg positive serum. First, Dane particles and the majority of the lipopro teins were precipitated from pooled sera by adding PEG 6000 to a final concentration of 5.5%. Then, HBsAg was precipitated out by adding PEG 6000 to the separated supernatant to a final concentration of 11%. The HBsAg pellet was dissolved in a buffer. Further purification was done by KBr buoyant-zonal density gradient centrifugation,and sucrose rate-zonal density gradient. The obtained HBsAg optic density unit of the remainder was 0.32%,i. e. 99.68% of other proteins were removed. The recovery rate of HBsAg was 73.4%. |
