摘    要:目的 :探讨乙肝疫苗接种后无、弱应答发生原因和机制。方法 :选择 2 1名乙肝疫苗接种后无、弱应答者为研究对象 ,同时设 2 2名强应答者为对照组。采用体外细胞培养和酶联免疫检测技术 ,分离外周血单个核细胞(PBMC) ,用特异性抗原HBsAg和非特异性抗原植物血凝素 (PHA)刺激培养 ,测定细胞培养上清中的白细胞介素 - 2(IL - 2 )及其可溶性受体 (sIL - 2R)水平。结果 :经HBsAg和PHA刺激后 ,无、弱应答组的PBMC培养上清中IL - 2水平明显低于强应答组 (P <0 .0 5 ) ,而sIL - 2R水平 2组间差异无统计学意义。结论 :PBMC分泌IL - 2水平低下可能是乙肝疫苗接种后无、弱应答发生的原因和机制之一。

Abstract:Aim: To study the mechanism of non-and-hyporesponse to Hepatitis B (HB) vaccine. Methods: ELISA was performed to determine the levels of IL-2 and sIL-2R in the supernatant of PBMC, which was isolated from the 22 hyperresponders and 21 non-and-hyporesponders and stimulated by HBsAg or PHA.Results:The levels of IL-2 in supernatants stimulated by HBsAg or PHA in non-and-hyporesponder group were lower than that in hyperresponder group( P < 0.05). The levels of sIL-2R in the supernatant stimulated by HBsAg or PHA had no significant difference ( P >0.5).Conclusion: Low levels of IL-2 production under the HBsAg or PHA stimulation may be one of the causes for non-and-hyporesponse to HB vaccine.
