胰淀素对大鼠葡萄糖和脂肪代谢的影响

摘    要:目的 :观察胰淀素对大鼠糖、脂肪代谢的影响。方法 :雄性SD大鼠 2 0只 ,随机分为 3组 ,以灌注生理盐水 (A组 )为对照 (n =7) ,灌注不同浓度 (B组 5nmol/h(n =7) ,C组 0 .5nmol/h(n =6 ) )的胰淀素 (amylin) 3h ,在灌注0min ,30min ,6 0min ,1 2 0min ,1 80min时采血测定游离脂肪酸 (NEFA)、甘油三酯 (TG)、胰岛素水平。后处死动物 ,取肝脏和肌肉以测定NEFA含量。结果 :灌注前A、B和C 3组动物体重、空腹血糖、胰岛素和血脂 (TG ,NEFA)各组间差异无统计学意义 (P >0 .0 5 )。灌注后B、C 2组血糖和胰岛素水平持续升高 ,30min时达到高峰 ,并分别高于A组 (P<0 .0 5 ) ;B和C组之间差异无统计学意义 (P >0 .0 5 )。B和C组灌注 6 0min时血NEFA开始升高 ,1 80min时升高最明显 ,并分别高于A组 (P <0 .0 5 ) ;B和C组之间差异无统计学意义 (P >0 .0 5 )。灌注 3h期间 ,A、B和C 3组血浆TG水平变化无统计学意义 (P >0 .0 5 )。肝脏NEFA含量在灌注 3h末 ,B和C组分别高于A组 ;3组肌肉NEFA含量差异无统计学意义 (P >0 .0 5 )。结论 :胰淀素能升高大鼠血糖和血NEFA水平

Abstract:Aim: To study the role of amylin on blood glucose and lipid metabolism in rats and its possible effect on inducing insulin resistance. Methods: Blood sample from fasting rats infused amylin(Group B 5 nmol/L,Group C 0.5 nmol/L) in different concentrations and control saline(Group A) were taken during infusing for detecting the levels of blood lipids, glucose, and insulin. Then, the rats were euthanized for determining the levels of NEFA, TG, and insulin in muscle and liver.Results: There were no differences in body weight, levels of fasting blood glucose, insulin, and blood lipid (triglyceride, TG, NEFA) among 3 groups. Plasma levels of glucose and insulin in Group B and C, which had a peak at 30 min, were higher than those in Group A during infusing amylin, respectively. But there was no difference in level of blood glucose between group B and C. Plasma NEFA levels in Group B and C, which were highest at 180 min, were higher than those in Group A from 60 min to 180 min during infusing amylin, respectively. But there was no difference of NEFA level between Group B and C. Plasma TG levels were unaltered during infusing amylin or saline among 3 groups. NEFA content of livers in Group B and C, but in muscle, was increased. Conclusion: Amylin has an effect on glucose and metabolism, which maybe induce insulin resistance.
