双波长分光光度法测定络石藤提取物总木脂素含量的专属性

摘    要:目的 考察已建立的双波长分光光度法测定络石藤提取物总木脂素含量方法的专属性。方法 以HPLC指纹图谱为检测方法,采用大孔树脂和聚酰胺两种柱层析方法对络石藤总木脂素提取物分别进行处理,使得到的两种产物的木脂素类成分和含量基本一致,但黄酮类成分和含量差异悬殊。而后采用双波长分光光度法对这两种产物进行总木脂素含量测定,若含量基本一致,表明分光光度法可以有效消除黄酮类成分对总木脂素含量测定的干扰,反之,需调整波长以提高专属性。结果 优化分离所得的大孔树脂层析产物和聚酰胺层析产物中木脂素类成分和含量基本一致,大孔树脂层析产物基本保留了络石藤总木脂素提取物中黄酮类成分,而聚酰胺层析产物中的黄酮类成分含量极低,并且两种产物的分光光度法测定所得的总木脂素含量基本一致,表明已建立的双波长分光光度法可以有效消除黄酮类成分的干扰。结论 已建立的紫外分光光度法测定络石藤提取物中总木脂素含量的方法具有良好的专属性。

Abstract:Objective To study the specificity of dual wavelength spectrophotometric quantitative analysis of total lignans in the extract from Trachelospermum jasminoides. Methods Detected by HPLC fingerprint analysis, the total lignans extract was chromatographied separately by macroporous resin and polyamide column to obtain two products in which the lignan components and their contents were extremely similar to each other but the flavonoid components and their contents were very different. Then total lignans contents in these two products were determined by the established dual wavelength spectrophotometry. If the two contents were closely equal, it′s sure that the interference from flavonoid components was well eliminated. On the contrary, if the two contents were very different, the wavelength should be adjusted to improve the specificity. Results Two optimized products in which the lignan components and their contents were extremely similar to each other were isolated. And the macroporous resin column chromatographied product had the similar flavonoid components to the total lignans extract, however polyamide column chromatographied product had very low contents of flavonoid components. Moreover, total lignan contents of these two products were closely equal. So, it was indicated that the dual wavelength spectrophotometry could well eliminate the interference from flavonoid components. Conclusion The established method for quantative analysis of total lignans in the lignan extract from Trachelospermum jasminoides by ultraviolet spectrophotometry has better specificity.
