野菊花反相高效液相色谱指纹图谱的建立及品质评价

摘    要:目的建立野菊花的反相高效液相色谱(RP-HPLC)指纹图谱和品质评价方法。方法采用优选的样品前处理方法,选用Kromasil C18(5μm,4.6 mm×250 mm)色谱柱,以乙腈-0.05%磷酸溶液为流动相(梯度洗脱),体积流量为1.0 ml/min,检测波长为334 nm,建立野菊花的RP-HPLC指纹图谱;以花形为小花,且相似度大于0.95,作为优质药材的判定标准,对22批不同产地野菊花进行品质评价。结果建立了野菊花的RP-HPLC指纹图谱,获得包含18个共有指纹峰的共有模式图谱,结合经验鉴别和指纹图谱建立了新的质量控制模式。结论本文建立的野菊花RP-HPLC指纹图谱方法专属性强,精密度和重复性良好,为野菊花整体质量控制提供了有效手段。

Abstract:Objective To establish a method of fingerprint analysis and quality evaluation on Flos Chrysanthemi Indici.Methods An optimized method of sample preparation was used,RP-HPLC was adopted with Kromasil C18(5 μm,4.6×250 mm),acetonitrile-0.05% H3PO4 solution(gradient elution) was used as a mobile phase with a flow rate of 1.0 ml/min,and detection wavelength was set at 254 nm.Fingerprint of 22 batches of Flos Chrysanthemi Indici from different regions were determined and their quality was evaluated: the good-quality flowers were floret and similarity between them was greater than 0.95.Results The fingerprint of Flos Chrysanthemi Indici by RP-HPLC was established,including 18 common peaks.Combined experiences identification with fingerprint to build new quality control mode.Conclusion With high specificity,precision and good repeatability,the established method provides an effective means for overall quality control of Flos Chrysanthemi Indici.
