| 摘 要： | 目的:研究重组人生长激素(rhGH)对裸鼠人胃癌移植瘤生长及血清VEGF水平的影响。方法:取60只雄性裸鼠进行实验,通过免疫细胞化学染色法检测胃癌细胞株SGC-7901及MKN-45生长激素受体(GHR)的表达状况,将GHR阳性表达的胃癌细胞株SGC-7901及GHR阴性表达的胃癌细胞株MKN-45分别接种于4只裸鼠皮下,制作裸鼠胃癌皮下移植瘤模型,3周后取出移植瘤制成组织匀浆,接种于剩余52只裸鼠。依据接种细胞种类不同分为移植瘤GHR阳性表达和GHR阴性表达裸鼠,不同GHR表达的裸鼠进一步分为对照组(C组:予0.9%NaCl,0.2 ml.d-1)、rhGH低剂量组(L组:予rhGH0.5 U.kg-1.d-1)、rhGH高剂量组(H组:予rhGH2.5 U.kg-1.d-1),连续给药14 d,观察并记录各组裸鼠体重和肿瘤体积变化,酶联免疫吸附法测定血清VEGF含量。结果:SGC-7901为GHR阳性表达的人胃癌细胞株,MKN-45为GHR阴性表达的人胃癌细胞株。对于接种GHR阳性表达的人胃癌细胞株的裸鼠,rhGH给药组皮下移植瘤体积较对照组增大(P<0.05),且H组促增长效应显著(P<0.05),3组...
