神经节苷脂GM1在颅脑损伤早期的脑保护作用

摘    要:目的:观察神经节苷脂GM1在急性颅脑损伤早期的脑保护作用.方法:在大鼠颅脑液压损伤后早期给予神经节苷脂GM1(30 mg*kg-1,伤后5 min和60 min各一次)或等量的生理盐水进行治疗,观察神经节苷脂GM1在伤后6 h对动物平均动脉压、脑水肿,以及损伤侧脑组织内乳酸和脂质过氧化物(LPO)的影响.结果:在颅脑液压损伤后,损伤组动物出现平均动脉压降低,损伤侧脑组织含水量、乳酸和LPO含量增加;而神经节苷脂GM1治疗组动物平均动脉压维持在伤前水平,损伤侧脑组织含水量、乳酸和LPO含量较损伤组动物显著下降(P<0.05或P<0.01),生理盐水对照组与颅脑损伤组之间无明显差异(P>0.05).结论:神经节苷脂GM1在颅脑损伤后早期有明显的脑保护作用.

Abstract:Objective: To observe the neuroprotective effects of ganglioside GM1 on the early traumatic brain injury(TBI). Methods: An acute experimental closed TBI model in rats was induced by a fluid percussion brain injury model. Fiveor 60 min after TBI, the animals were intraperitoneally injected with 30 mg' kg 1 of ganglioside GMl or same volumes ofsaline. The effects of ganglioside GM1 or saline on the changes of mean arterial pressure (MAP) and the contents of water,lactic acid (LA) and lipid peroxidation (LPO) in the injured cerebral tissues were observed 6 h after TBI. Results: AfterTBI, the MAP decreased and the contents of water, LA and l.PO increased in the brain injury group; However, the MAPmaintained at the pre-injured level and the contents of water, I.A and LPO decreased in the ganglioside GM1 treated groupwhen compared with the brain injury group (P<0. 05 or P<0. 01 ). There was no significant difference between the salinecontrol group and the brain injury group (P<0. 05). Conclusion: Ganglioside GM1 have obvious neuroprotective effect onthe early TBI.
