摘    要:目的:探讨去铁处理对实验性大鼠免疫性肝损伤的影响.方法:将48只Wistar大鼠随机分成单纯对照组、单纯肝损伤组、放血对照组、放血肝损伤组、去铁胺(DFO)对照组、DFO肝损伤组6组,通过放血或DFO疗法制做低铁模型,采用卡介苗加脂多糖诱导法复制免疫性肝损伤模型.检测血清铁(SI)、转铁蛋白(TRF)、总蛋白(TP)含量及天冬氨酸转氨酶(AST)活性,肝组织中丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性和肝铁含量(HIC)的变化.结果:与对照组比较,DFO对照组及放血对照组SI有降低趋势,但差异不显著;DFO对照组TRF增高,放血对照组TRF降低;肝损伤后SI水平和TRF均显著下降,AST均增高,TP均降低;DFO损伤组和放血损伤组AST活性增高幅度和TP含量的降低幅度均小于单纯肝损伤组.各组动物肝组织铁含量无显著差异.与对照组比较,DFO对照组及放血对照组MDA均显著降低,SOD活性仅有下降趋势.各损伤组与其相应对照组比较,MDA含量均增高,SOD活性均降低.结论:去铁处理降低铁负荷,可以改善肝损伤程度.

Abstract:Objective: To investigate the effect of reducing iron load on experiment immunological liver injury in rats. Methods: Totally 48 male Wistar rats were randomly divided into 6 groups:control group,liver injury group,phlebotomy control group,phlebotomy control group,phlebotomy liver injury group,deferoxamine control group(DFO),and deferoxamine liver injury group. The animal model of iron low load was made by DFO or phlebotomy. Immunological liver damage model was established by injection of lipopolysaccharide (LPS) and BCG (Bacilli Calmette Guein). Then the following parameters were determined: serum iron (SI), transferrin (TRF) concentration, total proteins (TP) volume, the serum activities of asparatate aminotransferase (AST), malondialdehyde (MDA) content, the activities of superoxide dismutase (SOD) and hepatic iron concentration(HIC) in liver tissue extracts. Results: In comparison with control group, the SI levels in DFO control and phlebotomy control groups only had a downward tendency. Compared with each own control groups, the serum activities of AST in all injury groups augmented and the TP levels reduced significantly. The increase of AST activities and the decrease of TP levels in DFO injury group and phlebotomy injury group were smaller than those of liver injury group. There was no significant difference between each injury groups and control groups concerning the hepatic iron load. In comparison with control group, the MDA levels in DFO control and phlebotomy control groups declined significantly and the activities of SOD in them had a downward tendency but with no significant difference. Compared with each own control group, the MDA levels were augmented and the activities of SOD reduced significantly in all injury groups. Conclusion: Iron low load can aggregate the severity of immunological liver injury in rats.
