摘    要:目的 构建霍乱毒素B亚单位编码基因(ctxB)的真核表达质粒pVAX1-ctxB,并观察其在真核细胞中的蛋白表达情况.方法 利用基因重组技术构建真核表达质粒pVAX1 -ctxB,通过脂质将其转染至CHO细胞中诱导蛋白表达,GM1 -ELISA法检测其蛋白表达产物.结果 重组质粒pVAX1-ctxB经酶切可得到与ctxB大小相同的片段,GM1 -ELISA法证实经转染的CHO细胞可分泌霍乱毒素B亚单位(ctxB)蛋白.结论 成功构建重组表达质粒pVAX1- ctxB,其蛋白表达产物具有ctxB蛋白活性,为下一步加强基因防龋疫苗效价奠定了基础.

Abstract:Objective:To evaluate the expression of recombinant plasmid pVAX1-ctxB in mammalian cell.Methods: The eukaryotic plasmid carrying encoding gene of ctxB was con-structed and introduced into CHO cells by lipofectamine reagent.The transient protein expression was detected with immunochemistry technique in CHO cells.Results: The positive expression of ctxB was detected in plasma of the cells which were transfected with recombinant plasmid pVAX1-ctxB.The cells which were transfected with pVAX1 were negative for ctxB expression.Conclusion: CtxB can be translated and expressed in CHO cells after transfected with recombinant plasmid pVAX1-ctxB.The expressed protein is located in the plasma to combine with the ctxB antibody.
