不同剂量腺嘌呤诱导肾阳虚不育大鼠模型实验研究

摘    要:目的探讨不同剂量腺嘌呤诱导肾阳虚不育大鼠模型的量效关系。方法将20只成年雄性SD大鼠随机分为4组:正常对照组,腺嘌呤低(每日100mg/kg)、中(每日150mg/kg)、高(每日200mg/kg)剂量组,每组5只。用腺嘌呤给予大鼠灌胃30d,诱导肾阳虚不育模型。观察大鼠体征、血清性激素水平、血尿素氮(blood urea nitrogen,BUN)、血清肌酐(serum creatinine,SCr)及睾丸、肾脏的形态学变化。结果与正常对照组相比,腺嘌呤各剂量组睾酮(testosterone,T)、雌二醇(estradiol,E2)显著降低(P〈0.05),卵泡刺激素(follicle stimulating hormone,FSH)、促黄体激素(luteotrophic hormone,LH)水平无显著性变化(P〉0.05),SCr、BUN水平显著升高(P〈0.05)。腺嘌呤各剂量组大鼠睾丸均出现生精小管中生精细胞排列紊乱,细胞疏松,精曲小管之间的结缔组织增多,间质细胞数量明显减少,管腔内生殖上皮变薄,精子数目减少,大部分管腔中可见脱落的精子细胞;肾小管内有大量结晶物沉着,且有异物肉芽肿形成,占据整个管腔,肾小管上皮肿胀、坏死、脱落,肾小管显著扩张成囊状,皮质、髓质间质纤维组织增生,大量炎性细胞浸润,可见纤维素样坏死。结论低剂量腺嘌呤可导致大鼠睾丸形态学改变,而且肾脏的损害程度较轻,可成功地诱导肾阳虚不育模型。
