摘    要:目的研究栀子苷(Jasminoidin,JA)在正常大鼠血浆中的药物代谢动力学和在肝、脾、肾、脑中的分布情况。方法采用phenomenex C18色谱柱(4.6 mm×250 mm,5μm),流动相为乙腈-0.1%磷酸(20∶80),流速0.8 ml/min,检测波长238 nm,柱温30℃。大鼠按JA单剂量60 mg/kg灌胃后,采用高效液相色谱法测定给药后不同时间点JA血药浓度和各组织浓度,通过DAS 2.0程序软件模拟计算,得出相应的药物代谢动力学参数。结果 JA的血药浓度-时间曲线符合权重为1的二室模型,各组织中药物浓度以肾脏最高,其他依次为肝、脾和脑。结论此方法简便、准确、稳定,可用于JA的药物代谢动力学和组织分布研究。

Abstract:Objective To study pharmacokinetics of Jasminoidin(JA) in rat plasma and distribution of JA in the liver,the spleen,the kidney and the brain of rats.Methods Phenomenex C18 column(4.6 mm×250 mm,5 μm) was used.The mobile phase was acetonitrile-0.01% phosphoric acid(20∶80);the flow rate was 0.8 ml/min;the column temperature was at 30 ℃;and the ultraviolet detection wave was set at 238 nm.High Performance Liquid Chromatography was used to study the concentration of JA and tissue distribution of JA in the brain,the liver,the spleen,and the kidney after intragastric administration of JA in normal rats at different time points.The pharmacokinetics parameters of JA were calculated by DAS 2.0 Program.Results The concentration-time curve of JA was fitted to a two-compartment model with a weight of 1.The concentration of JA was the highest in the liver,and it decreased in the liver,the spleen and the brain,gradually.Conclusion This method can be used for the study on pharmacokinetics and tissue distribution of JA because of its simplicity,accuracy and stability.
