红景天多糖对骨髓抑制小鼠表达凋亡相关蛋白的影响

摘    要:目的观察红景天多糖对骨髓抑制小鼠表达凋亡相关蛋白的影响,并探讨其造血调控作用部分机制。方法用免疫组化SABC法分别检测骨髓抑制小鼠骨髓细胞凋亡相关蛋白Caspases-3及Fas/FasL的表达。结暴经过射线照射及化疗药物造模后,小鼠骨髓细胞表达Fas/FasL、caspase-3明显增加,用药后各组小鼠骨髓细胞Fas/FasL、caspase-3蛋白表达有不同程度的减少,以高剂量组减少明显(P〈0.01)。结论红景天多糖可通过影响骨髓抑制小鼠骨髓细胞死亡受体Fas介导的Fas/FasL-caspase-3凋亡信号通路,减少骨髓细胞凋亡,促进骨髓造血功能的恢复。

Abstract:Objective: To observe the effects of Rhodiola polysaccharide on apoptosis related protein expression of rats with myelosuppression, and to investigate the mechanism of hematopoietic regulation. Methods: Immunohistochemical SABC method was employed to detect the expression of myelosuppression rats' apoptosis related protein Caspases-3 and Fas/FasL. Results: After irradiation and chemotherapy, rats'bone marrow cells'expression of Caspases -3 and Fas/FasL increased obviously. After giving the medicine, the expression of Caspases-3 and Fas/FasL of each group decreased in various degrees, and the decrease in the high dosage group was the most obvious (P〈0.01). Conclusion: Rhodiola poly- saccharide can reduce the apoptosis of bone marrow ceils and promote the recovery of its hemopoietic function by affecting Fas/FasL- caspase-3 apoptosis signal pathway mediated by myelosurression rats'bone marrow cell death receptor Fas.
