保肝护脑针法治疗脑缺血再灌注损伤的实验研究

摘    要:目的探讨保肝护脑针法对脑缺血再灌注大鼠的治疗作用。方法成年SD雄性大鼠40只,随机分为假手术组、模型组、针刺对照组、保肝护脑针法组,每组10只。制作大脑中动脉阻断脑缺血大鼠模型,测试大鼠行为学、检测血清谷丙转氨酶(ALT)、超氧化物歧化酶(SOD)、丙二醛(MDA)。结果模型组、针刺对照组和保肝护脑针法组缺血2 h(电针治疗前)时神经功能缺损评分无显著差异;经10次电针治疗结束后:与模型组相比,针刺对照组和保肝护脑针法组神经功能缺损评分有极显著差异(P<0.01);与针刺对照组相比,保肝护脑针法组神经功能缺损评分变化更为显著(P<0.05)。电针治疗前,与假手术组、针刺对照组和保肝护脑针法组比较,模型组血清ALT、MDA水平显著增高(P<0.01),而血清SOD活性明显降低(P<0.01);经10次电针治疗结束后:与模型组相比,针刺对照组和保肝护脑针法组血清ALT、MDA水平明显降低(P<0.01),同时血清SOD活性显著升高(P<0.01);与针刺对照组相比,保肝护脑针法组血清ALT、SOD、MDA各项变化更为显著(P<0.05)。结论保肝护脑针法对脑缺血再灌注大鼠的治疗效果较单纯头穴针刺更为显著,其治疗机理与抗氧应激密切相关。

Abstract:Objective To discuss the therapeutic effect of cerebral ischemia reperfusion injury with acupuncture therapy of protecting liver and brain.Methods 40 male adult SD rats were randomly and evenly divided into four groups: the model group,the sham operated group,the acupuncture control group and the acupuncture therapy of protecting liver and brain group.Firstly a rat model with cerebral ischemia caused by middle cerebral artery occlusion(MCAO) was made.Then,rats’ ethological behaviors were tested.At last,serum alanine transaminase(ALT),superoxide dismutase(SOD) and malondialdehyde(MDA) were detected.Results Neurologic deficit scores(NDS) were not significantly different among the three groups when cerebral ischemia lasted for two hours(before electro-acupuncture therapy).After performing electro-acupuncture therapy for ten times,compared with the model group,NDS are greatly different(P<0.01) between the acupuncture control group and the acupuncture therapy of protecting liver and brain group.And,compared with the acupuncture control group,NDS changed a lot(P<0.05) in the acupuncture therapy of protecting liver and brain group.Before electro-acupuncture therapy,compared with the sham operated group,the acupuncture control group and the acupuncture therapy of protecting liver and brain group,the level of serum ALT and MDA in the model group increased significantly(P<0.01),however,the activity of SOD decreased significantly(P<0.01);After performing electro-acupuncture therapy for ten times,compared with the model group,the level of serum ALT and MDA in the acupuncture control group and the acupuncture therapy of protecting liver and brain group decreased significantly(P<0.01),however,the activity of serum SOD increased significantly(P<0.01).Compared with the acupuncture control group,serum ALT,SOD and MDA in the acupuncture therapy of protecting liver and brain group changed significantly(P<0.05).Conclusion Comparing with simply needling on scalp points,protecting liver and brain therapy is more effective when treating rats with cerebral ischemia reperfusion and its therapeutic mechanism is closely related with antioxidant stress.
