针刺乳腺增生大鼠的乳腺形态学改变

目的通过系统观察乳腺增生大鼠及针刺治疗后乳腺组织形态学变化过程,探讨针刺对乳腺增生病的疗效与作用机理,为临床针刺治疗乳腺增生提供可靠的形态学依据。方法将40只健康雌性大白鼠随机分为4组,每组10只:A空白对照组、B模型组、C自愈组和D针刺治疗组。除A组外均采用饶氏造模方法建立乳腺增生模型。B、C两组大鼠于第26 d开始每日抓拿一次,不作任何治疗;D组大鼠在第26 d开始针刺治疗。治疗30 d后处死大鼠,游标卡尺观测乳房形态大小后,采集乳腺组织标本,分别固定后染色,光镜下观察其结构的病理变化。结果镜下结构显示造模成功,细胞结构改变明显;针刺治疗组乳腺组织病理改变明显轻于模型组,趋近于空白对照组。结论针刺对乳腺增生大鼠乳腺组织有抑制作用,说明针刺对乳腺增生病有治疗作用。

Morphological Change of Mammary Gland of Rats with Mammary Gland Hyperplasia after Needling

Objective: By systematically observe rats with mammary gland hyperplasia and morphological change of the tissue of mammary gland after needling,to probe into the efficacy and mechanism of needling on mammary gland hyperplasia,and to provide reliable morphological evidence for needling in the treatment of mammary gland hyperplasia.Methods: 40 healthy female white rats were randomly divided into 4 groups,10 in each group: blank control group(A),model group(B),natural cure group(C) and needling group(D).All groups were built mammary gland hyperplasia model by Rao Shi model-building method except blank control group.The rats in model group,natural cure group and needling group were injected with estradiol benzoate through muscles at lateral of posterior limbs for 20 days,0.5mg/kg weight,1time per day,then changed to progesterone for 5 days,5mg/kg weight,1 time per day.The blank control group were injected with equal amount 0.9% normal saline,1 time per day,25 days altogether.The rats in group B and C were caught one time per day from the 26th day with no treatment.The rats in group D were treated with needling from the 26th day.The rats were killed after 30 days of treatment.Using vernier to observe the morphology and size of the breast.Collecting the samples of mammary gland tissues to fix and stain.Observing its pathological change under opticalmicroscope.Results: The experimental results show that the model building in model group was successful,and the cell structure was changed greatly;the pathological change of mammary glands of needling group was obviously less obvious than the model group,which is close to the blank group.Conclusion: Needling has an inhibitory effect on the mammary gland tissues of rats with mammary gland hyperplasua,which indicates that needling is effective for the treatment of mammary gland hyperplasia.
