| Abstract: | Objective] To investigate the expression of GRP94 under A23187 and BAPTA-AM and its function in the resistance to cisplatin in a human lung cancer NCI-H460 cell line.Methods] The cells were devided into 3 groups: A23187,BAPTA-AM and control.RT-PCR and western blot were used to detected the expressions of GRP94 at mRNA and protein levels in 3 groups.MTT was used to determine the effect of cisplation on cell survival.Results] The expressions of GRP94 at both mRNA and protein levels were increased obviously in NCI-H460 cell line induced by A23187,but decreased obviously in NCI-H460 cell line inhibited by BAPTA-AM,and MTT assay showed that the IC50 of A23187,BAPTA-AM and control groups to cisplatin were 3.34±0.12,4.82±0.29 and 8.12±0.21,respectively(P<0.05).Conclusion] The expression of GRP94 might enhance the sensitivity of NCI-H460 cell line to cisplatin. |
