血管紧张素-(1-7)对TGF-β1诱导的大鼠心肌成纤维细胞Smad3、Smad7 mRNA表达的影响

摘    要:目的:观察血管紧张素-(1—7)对TGF-β1诱导幼年大鼠心肌成纤维细胞增殖以及Smad3、Smad7mRNA表达的影响。方法:分离培养SD仔鼠心脏成纤维细胞,以Ang-(1—7)、TGF-β1、Ang-(1—7)+TGF-β1组、TGF-βI型受体抑制剂等干预,通过WST-1法检测细胞增殖,RT—PCR测定心脏成纤维细胞Smad3、Smad7mRNA的表达。结果:①与空白对照组比较,TGF-β1组心脏成纤维细胞的OD值增加(P〈0.05);与TGF-β1组比较,0.001~1.0μmol/L血管紧张素-(1-7)和5μg/TGF-β1共同干预后OD值逐渐降低(P〈0.05)。②与对照组比较,TGF-β1组心脏成纤维细胞的Smad3mRNA表达升高(P〈0.05),Smad7mRNA表达降低(P〈0.05)。结论:血管紧张素-(1—7)能有效抑制TGF-β1引起的心脏成纤维细胞的增殖,其作用不通过TGF-βI型受体介导。

EFFECT OF ANGIOTENSIN-(1-7)ON SMAD3 AND SMAD7 MRNA EXPRESSION OF RAT CARDIAC FIBROBLASTS INDUCED BY TGF-β1

Abstract:Objective: To investigate the effect of angiotensin -(1-7) Ang-(1-7)] on cell proliferation and the expression of Smad3 and Smad7 mRNA of cardiac fibroblasts (CF) induced by TGF-β1. Methods: CF were isolated and cultured from Neonatal SD rats.The cells were divided into different groups and treated with Ang-(1- 7), TGF-v1, TGF-β1+ Ang-(1-7), and the acceptor inhibiter of TGF-β1 respectively. Cell number was evaluated by WST-1. Smad3 and Smad7 mRNA expression in CF were analyzed by RT-PCR. Results: ①Compared with the control group, 5μg/L TGF-β1 increased absorption value of CF(P〈0.05). Compared with 5μg/L TGF-β1 group, TGF-β1+ 0.001 -1.0μmol/L Ang-(1-7) attenuated the absorption value of CF in a concentration dependent manner (P〈0.05). ②Compared with TGF-β1 group, the expression of Smad3 mRNA decreased, while the expression of Smad7 mRNA increased in TGF-β1+Ang-(1-7) group (P〈0.05). Conclusion: Ang-(1-7) can downregulate the expression of Smad3 mRNA, and up-regulate the expression of Smad7 mRNA induced by TGF-β1.
