小檗碱在抑制兔颈动脉粥样硬化形成中的作用

摘    要:目的:探讨小檗碱抑制家兔颈动脉粥样硬化形成的作用机制。方法:24只雄性新西兰大白兔随机分为3组。正常对照组予普通饮食5周;模型组予高脂饲料喂养1周后,行右侧颈动脉内膜空气干燥术,再高脂喂养4周制作颈动脉粥样硬化模型;小檗碱处理组每日同时予颈动脉粥样硬化模型兔灌服小檗碱。观察颈动脉病理改变,并检测组织中巨噬细胞沉积和核因子-кB(NF-кB)、单核细胞趋化蛋白-1(MCP-1)表达情况。结果:模型组颈动脉有明显粥样硬化斑块形成,可见大量巨噬细胞沉积,NF-кB p65和MCP-1蛋白及mRNA表达明显(P<0.05),小檗碱组颈动脉硬化程度较轻,巨噬细胞明显减少,NF-кB p65和MCP-1蛋白及mRNA表达较模型组低(P<0.05),NF-кB与MCP-1二者呈正相关(r=0.958,P<0.01)。结论:小檗碱可能通过抑制NF-кB活性、降低MCP-1表达,减少巨噬细胞沉积而干预颈动脉粥样硬化形成。
