E1B缺陷腺病毒dl1520预感染CNE-2细胞对其致瘤性的影响

目的探讨E1B-55KDa缺陷腺病毒dl1520预感染鼻咽癌细胞CNE-2对其致瘤性的影响。方法10MOI(感染复数)的dl1520感染CNE-2细胞后,按5%、20%的比例将感染细胞和未感染细胞混合后做裸鼠皮下注射,观察肿瘤生长情况并采用免疫组化检测腺病毒六邻体抗原,分析dl1520在肿瘤内的复制及分布情况。结果5%、20%组均可见肿瘤生长明显抑制,抑瘤率分别为30.59%、76.60%,P<0.01。免疫组化证实瘤内有大量病毒复制且20%组比5%组分布更广泛。结论dl1520预感染CNE-2细胞后对其裸鼠移植瘤的生长有明显抑制作用,进一步证实dl1520具有一定的抗肿瘤作用且病毒在瘤内的分布情况可影响抗肿瘤的效果。

The effect of E1B-deleted Adenovirus dl1520 on the Tumorigenicity of preinfected Nasopharyngeal Carcinoma cell CNE-2

Objective To evaluate the tumorigenicity of nasopharyngeal carcinoma cell CNE-2 preinfected with E1B-deleted adenovirus dl1520. Methods CNE-2 were infected with dl1520 at a 10 multiplicity of infection(MOI) for 3 hours and mixed with uninfected cell suspension at 5% and 20% rations;then cells were injected into the flanks of nude mice. The tumor growth was monitored and adenovirus hexon protein was detected by immunohistochemistry to evaluate propagation and distribution of dl1520 in tumor. Results In 5% and 20% groups tumor growth inhibition was observed obviously; inhibition rate was 30.59%、76.60%(P<0.01), respectively. Immunohistochemistry showed that distribution of dl1520 was more widely in 20% group than in 5% group. Conclusion Dl1520 can significantly inhibit the tumorigenicity of preinfected tumor cell; this further confirmed that dl1520 has anti-tumor effect which can be influenced by its distribution in tumor.
