| 摘 要： | 目的 研究特异性细胞毒Ｔ淋巴细胞杀伤胃癌细胞ＢＧＣ－８２３的超微结构变化。方法 用２２例患者的胃腺癌细胞反复冻融后的滤液，抗ＣＤ３单克隆抗体、白细胞介素－２（ＩＬ－２）患者自体外周血淋巴细胞共同培养，体外诱导ＣＴＬ，然后将ＣＴＬ与ＢＧＣ－８２３共同孵育，用电子显微镜观察ＣＴＬ作用于ＢＧＣ－８２３后的超微结构改变。结果 电镜观察可见ＣＴＬ包围并破坏靶细胞，靶细胞凋亡的形态表现为异染色质浓缩呈特征性半 |

| Abstract: | ObjectiveTo study ultrastructural changes of BGC 823 gastric cancer cells due to the killing effects of the specific cytotoxic T lymphocytes(CTL).MethodsThe CTLs were induced by repeated freeze thawed extracts of gastric cancer cells from 22 patients with gastric carcinoma and anti CD 3 monoclonal antibody as well as recombinant interleukin 2 in vitro.The CTLs were then cultured with target cells BGC 823.Ultrastrutural changes of BGC 823 cells after CTL treatment were observed with electron microscope.ResultsThe target cells were frequently surrounded and attacked by CTLs.Apoptotic changes,such as heterochromatin condensation with characteristic crescentic cap appearance and necrostic changes were found in BGC 823 cells.ConclusionThe specific CTL killed target cells by inducing apoptosis and necrosis. |
