组织工程化表皮膜片修复裸鼠创面的实验研究

摘    要:目的 构建组织工程化表皮细胞膜片并应用于修复裸鼠全层皮肤缺损创面。方法 体外分离、培养、扩增人类表皮细胞,与壳聚糖—明胶膜构建表皮细胞膜片,用于修复裸鼠背部全层皮肤缺损创面,以单纯壳聚糖/明胶膜作为对照;术后行大体、组织学观察及免疫组织化学检测。结果 表皮细胞可以壳聚精—明胶膜作为载体在体外培养,达到60%~70%亚融合状态时,移植人表皮细胞/壳聚糖—明胶膜到裸鼠全层皮肤缺损创面,人表皮细胞可以在创面存活并继续增殖分化,形成连续的表皮覆盖创面。结论 人表皮细胞/壳聚糖—明胶膜是一种理想的组织工程表皮替代物。

Abstract:Objective To construct tissue engineered epidermal membrane and explore the possibility of repairing the full-thickness skin defects of nude mouse with engineered membrane. Methods Seeded the cultured epidermal cells (EC) on chitosan-gelatin(CG) membrane to form an artificial epidermal grafts. Then the engineered membrane was transplanted to repair the full-thickness skin defects of nude mouse. CG membrane without EC served as control. Results Epidermal cells could be cultured in vitro on CG membrane. When EC reached 60%-70% sub-confluence , the EC-CG membrane was grafted. The human EC could be survived on the wound and continued to proliferate and differentiate to form a layer of new epidermis. Conclusion EC-CG membrane may become an ideal tissue engineered epidermal subsittute.
