激光诱导兔脉络膜视网膜静脉吻合中VEGF水平的变化

摘    要:目的 研究激光诱导脉络膜视网膜静脉吻合(CRVA)形成过程中血管内皮生长因子(VEGF)水平的变化,探讨VEGF与CRVA形成的关系。方法 以激光诱导兔CRVA的常规方法对兔眼行激光处理,在激光术后3 d和1、2、4、6周ELISA法检测实验兔眼房水、玻璃体、激光击射部位和非击射处视网膜脉络膜组织的VEGF含量。以正常兔眼及单纯-过性静脉阻塞非CRVA激光处理组(RVO组)作为对照。结果 正常兔眼含有较低水平VEGF。RVO组和CRVA组均于激光术后1周出现VEGF峰值,两组峰值比较无明显差异(P>0.05);术后2~4周cRVA组仍保持较高水平VEGF,与同期RVO组相比差异显著(P<0.05)。结论 CRVA组VEGF峰值维持至激光术后4周,与CRVA形成时间相吻合,推测VEGF水平的升高与CRVA的形成过程有一定的内在联系。

Abstract:Objective To investigate the content of vascular endothelial growth factor (VEGF) in rabbit eyes after laser-induced chorioretinal venous anastomosis (CRVA) , in order to find out the relationship between CRVA and VEGF. Methods Undiluted aqueous humor, vitreous and diluted homogenate of the retina and choroids were obtained in rabbit eyes after laser-induced chorioretinal venous anastomosis on the basis of retinal vein occlusion (RVO) , which has been established in our laboratory. VEGF levels were detected by using enzyme linked immu-nosorbent assay (ELISA) in group CRVA and group RVO, respectively in 3 d and 1,2,4,6 weeks after laser. Results In the noxmal group, the mean value of VEGF was low. The VEGF levels were significantly higher in group CRVA and group RVO one week after laser, but there was no significant difference between the two groups ( P > 0. 05). There were higher levels in the group CRVA between two and four weeks after laser. In comparison with the RVO group, the difference was statistically significant at the same time (P <0.05). Conclusion VEGF keeps higher levels in the group CRVA in four weeks after laser, and coincides with laser-induced CRVA. VEGF plays a role in the neovascularization of CRVA.
