紫杉醇耐药与敏感细胞的差异表达蛋白分析

摘    要:目的 比较人肺腺癌细胞系A549与肺腺癌紫杉醇耐药细胞系A549-Taxol的差异表达蛋白.方法 提取A549和A549-Taxol总蛋白,应用双向凝胶电泳技术(2-DE)分离,ImageMaster 5.0软件分析并筛选出两者间差异表达的蛋白点,质谱分析加以鉴定.结果 获得分辨率高且重复性好的肺腺癌耐紫杉醇细胞系及亲代细胞系2-DE图谱.ImageMaster软件分析差异大于2倍的蛋白质点共有80个,其中33个在耐药细胞系中上调,47个下调.质谱分析鉴定出19种差异表达蛋白,其中A549-Taxol中上调蛋白11种,涉及细胞骨架蛋白、代谢酶类、分子伴侣和信号转导相关蛋白质.结论 应用2-DE技术整体展示了紫杉醇耐药细胞系及敏感细胞系的蛋白表达谱,质谱法鉴定的差异蛋白为进一步从多因素角度研究紫杉醇耐药机制提供了新线索.

Abstract:Objective To compare the protein expression profiles between human lung adenocarcinoma cell line A549 and paclitaxel resistant cell line A549-Taxol.
