可溶性白细胞介素-4受体在哮喘小鼠肺组织中的变化及地塞米松的干预作用

摘    要:目的 探讨可溶性白细胞介素 4受体 (sIL 4R)在哮喘发病中的作用及地塞米松(DXM)对sIL 4R的影响。方法 用卵清白蛋白复制BALB/c小鼠哮喘模型 ,RT PCR方法检测哮喘小鼠基础的和DXM干预后肺组织sIL 4RmRNA的表达变化。结果 哮喘小鼠支气管管壁及血管周围大量炎性细胞浸润 ,以嗜酸性粒细胞 (EOS)为主。经DXM干预后肺组织中炎性细胞浸润明显减少。哮喘组小鼠的sIL 4RmRNA相对丰度显著下调。经DXM干预后 ,sIL 4R基因相对丰度显著增高。结论 支气管哮喘急性发作时 ,存在sIL 4RmRNA表达下调 ;DXM可纠正sIL 4RmRNA的表达下调。

Abstract:Objective To elucidate the role of soluble interleukin 4 receptor (sIL-4R) in pathophysiology of asthma and the effect of dexamethasone (DXM) on its expression.Methods Asthma model of BALB/c mouse was established by administration of OVA as an allergen.The sIL-4R mRNA from the asthmatic group before and after intervention of DXM was detected by RT-PCR.Results Murine model of atopic asthma was successfully copied.Histological evaluation of pulmonary tissue demonstrated characteristic pulmonary eosinophilic inflammation which could be dramatically suppressed by DXM.The abundance of sIL-4R mRNA in OVA challenged mice was lower than that in control groups.DXM may up-regulate the expression of sIL-4R gene.Conclusions Down-regulation of sIL-4R gene may contribute to asthma pathogenesis and up-regulation of sIL-4R expression might be one of the therapeutic mechanisms of DXM.
