摘    要:目的 探讨特异性免疫治疗(SIT)对哮喘大鼠模型白细胞介素-10(IL-10)和转化生长因子-β1(TGF-β1)的影响.方法 40只健康雄性清洁级Wistar大鼠随机分成正常对照组(C组)、哮喘组(A组)、SIT对照组(CT组)和SIT治疗组(T组),各10只.通过卵蛋白(OVA)雾化吸入的方法对致敏大鼠进行SIT干预,观察各组支气管肺泡灌洗液(BALF)中细胞分类及计数结果、血清和BALF中IL-10及TGF-β1水平变化.结果 C组BALF和血清中IL-10浓度分别高于A组和CT组〈P<0.01),A组BALF和血清中IL-10浓度分别低于T组(P〈0.01):A组血清和BALF中TGF-β1水平分别高于C组(P〈0.01)和T组(P〈0.05,P〈0.01),T组血清和BALF中TGF-β1水平均分别高于C组(P〈0.05).结论 通过调节体内IL-10和TGF-β1趋于正常状态可能是SIT治疗哮喘有效的重要机制.

Abstract:Objective To investigate the effect of specific immunotherapy on IL-10 and TGF-β1 levels in serum and bronchoalveolar lavage fluids (BALF) of asthmatic rats. Methods Forty Wistar rats were randomly divided into 4 groups (n=10 in each group): the control group(group C),the asthma group(group A), the control of specific immunotherapy group(group CT) and the specific immunotherapy group(group T). The experimental asthma was established by using ovalbumin (OVA) challenge methods and the rats in group T were received the repeated inhaling different doses of OVA. The percentage of cells in BALF, the concentration of IL-10 and TGF-β1 in BALF and serum were measured respectively. Results The concentrations of IL-10 in BALF and serum of group A were significantly lower than those of group C( 0.01)or group T( 0.01)respectively. The serum IL-10 level in the group T was also lower than that of group C( 0.05). TGF-β1 levels in BALF and serum of group A were significantly higher than those of group C ( 0.01)or group T( 0.05)respectively. TGF-β1 levels in BALF and serum of group C was significantly lower those of group T( 0.05). Conclusions The data suggested that the specific immunotherapy could prevent the airway inflammation by increasing IL-10 and reducing TGF-β1 in asthmatic rat.
