摘    要:目的:研究血管内皮生长因子(VEGF),基质金属蛋白酶-2(MMP-2)在非小细胞肺癌(NSCLC)中的表达及其与肿瘤血管形成的关系。方法:用免疫组化(S-P法),染色技术对95例NSCLC石蜡组织标本的VEGF、MMP-2及肿瘤微血管密度(MVD)进行检测和分析。结果:VEGF、MMP-2在肿瘤细胞阳性表达率明显高于癌旁的正常肺组织(P<0.01)。VEGF及MMP-2阳性表达癌组织中的MVD显著高于阴性表达癌组织中的MVD。VEGF表达与TNM分期及淋巴结转移有关(P<0.05),和肿瘤病理分级增高无显著相关(P>0.05)。MMP-2表达与肿瘤病理分级及淋巴结转移有关(P<0.05)。结论:VEGF和MMP-2可能参与了NSCLC的新血管生成,故VEGF、MMP-2的检测可作为判断NSCLC转移及预后的指标。

Abstract:Objectives: To investigate the expression and distribution of VEGF, MMP 2 in human non small cell lung cancer (NSCLC),and to evaluate the relationship between the expressions of VEGF and MMP 2 and tumor angiogenesis and metastasis. Methods:The expression of VEGF, MMP 2 and MVD in 95 patients with NSCLC were examined using immunohistochemical methods (S P), and the relationships between them and many clinicopathological parameters were analyzed. Results:The positive rates of VEGF and MMP 2 in NSCLC were much higher than those in the normal lung tissues nearby ( P < 0.01 ). The positive expressions of VEGF and MMP 2 were associated with MVD and lymph node metastases of NSCLC( P <0.05). The positive expression of VEGF was associated with TNM stage ( P < 0.01 ). Conclusions: The results suggested that VEGF, MMP 2 may play an important role in tumor angiogenesis and metastasis of human NSCLC, and detection of VEGF and MMP 2 may be good metastasis and prognostic predictors for human NSCLC.
