摘    要:目的:观察汉坦病毒(HV)体外感染非洲绿猴肾上皮细胞(Vero-E6细胞)诱导热休克蛋白(HSP)的表达并探讨其与病毒结构蛋白的相互关系。方法:采用HV76-118株体外实验性感染Vero-E6细胞,随机分为对照组和病毒感染组,运用免疫细胞化学染色、Western blot及双特异性抗体夹心酶联免疫吸附法(ELISA),观察HV感染后HSP27、HSP70、葡萄糖调节蛋白(Grp94)3种HSP的表达情况,并分析上述HSP与病毒核衣壳蛋白(NP)之间的相互关系。结果:HV76-118株感染Vero-E6细胞24 h后,免疫细胞化学染色法可检出HV-NP阳性信号。Westernblot结果表明,与对照组相比较,HSP27、HSP70、Grp94的表达水平在感染后24 h显著升高(P〈0.05,n=5)。更为重要的是,ELISA检测发现HV-NP同时与HSP27、HSP70、Grp94呈复合物形式存在。结论:HV体外感染可诱导Vero-E6细胞高表达多种HSP,且能与病毒结构蛋白形成复合物,上述过程可能参与了HV-宿主细胞的相互作用。

Abstract:Objective:To study heat shock proteins(HSP) and their association with Hantavirus nucleocapsid protein(HV-NP) in Vero-E6 cells infected with Hantaan76-118.Methods: The HV-NP was identified by immunocytochemical staining after infected with Hantaan76-118.The expression of HSP was detected by Western-blot and analyzed by double specific antibody sandwich ELISA.Results: Western-blot exhibited that the expressions of HSP27,HSP70 and Grp94 in the Vero-E6 cells infected with Hantaan 76-118 were significantly higher than those in the control cells(P<0.05,n=5).More importantly,HSP27,HSP70 and Grp94 were associated with HV-NP(n=5), which led to the formation of protein complexes.Conclusion: HV infection induces the expression of a set of HSP including HSP27,HSP70 and Grp94,which are associated with HV-NP.This association may demonstrate the chaperone role of HSP in the hantavirus-host interaction.
