摘    要:目的:改进制备工艺,分离、纯化东海近江牡蛎(Ostrea rivularis Gould)得到牡蛎多糖(Oyster polysaccharides,OPs),并探索其生物学活性。方法:采用热碱法提取,以三个剂量组(2.5、5、10mg/ml)进行正常小鼠脾淋巴细胞转化实验,迟发型超敏反应(DTH)、自然杀伤细胞(NK)活性测定,抗体形成细胞活性溶血空斑形成细胞实验(PFC)、碳廓清实验、腹腔巨噬细胞吞噬鸡红细胞实验、荷瘤小鼠脾淋巴细胞活性测定(MTT法)、红细胞超氧化物歧化酶(SOD)活性测定。结果:制备得到牡蛎多糖粗品;免疫学实验表明OPs2.5mg/ml组能有效增加正常小鼠的脾淋巴细胞转化率、对小鼠DHT、NK细胞活性、PFC、小鼠腹腔巨噬细胞吞噬鸡红细胞能力、荷瘤小鼠脾细胞活性起正向调节。三种浓度实验组均增强小鼠红细胞SOD活性。结论:牡蛎多糖能增强小鼠细胞免疫、体液免疫功能,并有一定的抗肿瘤和抗氧化作用。

Abstract:Objective: We extracted and purified polysaccharides from Ostrea rivularis in east China sea,and studied the biological activity of oyster polysaccharides on the immune system. Methods: To study its biological activity,crude OPs were extracted and purified,and three dosages were applied in various methods such as mouse spleen lymphocytes transformation test(LTT),delayed type hypersensitivity(DTH)reaction,test of plaque forming cell,NK cell activity assay,assay of phagocytic function of the phagocyte,tumor-bearing mouse spleen lymphocyte activity test and assay of erythrocytic SOD activity.Results: Comparing with the control group,the purified crude OPs(2.5 mg/ml) could significantly improve the function or activity of mouse spleen lymphocytes transformation,plaque forming cell,NK cell,phagocyte,tumor-bearing mouse spleen lymphocyte,while remarkably improved the effect of DTH.Additionally,the activity of erythrocytic SOD were improved with respective dosage. Conclusion: OPs can facilitate several immunological functions and activity of anti-tumor and anti-oxidation.
