利用多重荧光STR技术分析上海地区7品系常用近交系小鼠核心群的遗传特性

目的 分析比较上海地区7品系常用近交系小鼠核心群的遗传特性。 方法 将筛选到的48对多态性丰富的微卫星引物组合优化，形成11组多重荧光PCR引物混合体系，对来自上海地区两大实验小鼠供应商的7品系14种群（亚系）近交系小鼠核心群的DNA样进行分型检测。利用遗传分析软件进行数据分析。结果 14种群（亚系）近交系小鼠在48个微卫星位点上都为纯合子，同品系小鼠在同一种群(亚系)内表现为单态性，在不同种群(亚系)间存在差异；但同品系不同种群(亚系)近交系小鼠间的遗传距离与品系间相比均较近，均首先两两聚成一类。C57BL/6小鼠与其他6品系小鼠的亲缘关系均较远。结论 上海地区不同供应商的同品系近交系小鼠，不论是否为相同亚系，其遗传差异均存在。

Genetic Characteristics of Seven Inbred Mouse Strains in Core Colonys from Shanghai Analyzed by the Multiplex Fluorescence STR Technique

Objective To analyze the genetic characteristics of seven frequently-used inbred mouse strains in core colonys from Shanghai Suppliers. Methods 48 pairs of microsatellite primers with high polymorphism were screened out and combined to 11 sets of multiplex PCR primers mix labelled with two dyes (FAM and HEX). By these primer mix, the DNA samples of the seven strains -fourteen populations (substrains) were genotyped. The genetyping datas were analyzed by statistical software. Results The 48 microsatellite loci were all homozygous for all inbred mice of the fourteen populations (substrains). On the same locus, the PCR results of the same strain mice showed monomorphism in the same populations (substrains), and showed polymorphism in different populations (substrains). But the genetic distance between the populations (substrains) of the same strains were all smaller than that of the different strains. The phylogenic tree showed the populations (substrains) of the same strain were first get into one cluster. The genetic relationship between the strain of C57BL/6 and other six strains were all far. Conclusion The genetic difference of the same strain from two different Suppliers in Shanghai were existing whether the populations were the same subtrain or not.
