不同浓度和纯度盐酸小檗碱大鼠肠道吸收动力学比较研究

摘    要:目的:考察盐酸小檗碱在大鼠各肠段的吸收动力学特征及不同药物浓度和纯度对大鼠小肠吸收动力学的影响。方法:利用大鼠在体肠吸收实验模型,分别采用HPLC法和UV法测定肠循环液中盐酸小檗碱和酚红的浓度,对盐酸小檗碱大鼠在体肠吸收特性进行研究。结果:盐酸小檗碱在肠道中的吸收按十二指肠、空肠、回肠的顺序依次下降;随着盐酸小檗碱浓度的增加,吸收速率常数(Ka)基本保持不变,吸收百分率(P)逐渐增加;且低纯度药物吸收百分率(P)显著高于高纯度药物(P<0.05)。结论:盐酸小檗碱在十二指肠、空肠吸收较好,其机制为被动扩散;低纯度的药物吸收较好。

Study on absorption of berberine of different concentratioils and purities in rat's intestine
Abstract:Objective: To investigate the absorption kinetics of berberine in different intestinal segments of rats and the influence of different purites and concentrations of berberine in rats intestine.Methods:In situ pass perfusion model and HPLC method was used to determine the concection of berberine the perfusion which outleted from the opened rat in vivo.Results:The absorption of berberine decreased according to the turn of ileum,jejunum and duodenum.With the increase in concentration of berberine P increased,and P of the lower purity of berberine higher than the higher purity.Conclusion: Berberine can be absorbed in intestinal segments,and absoption mechanism was passive diffusion,the lower purity of berberine has better absorption.
