和厚朴酚对人宫颈癌细胞增殖和凋亡的影响

摘    要:目的探讨和厚朴酚抑制人宫颈癌Hela细胞增殖及诱导其凋亡的作用。方法利用MTT检测和厚朴酚在不同浓度和时间下对Hela细胞增殖的影响;用流式细胞术、Hoechst 33258荧光染色和DNA ladder检测和厚朴酚作用下Hela细胞的凋亡。结果MTT结果显示和厚朴酚可明显抑制Hela细胞的增殖,且抑制率存在剂量和时间依赖性;流式结果表明10μg/mL和20μg/mL和厚朴酚作用细胞24h后,凋亡细胞的比例分别为22.5%和62.2%,而对照组为8.7%;和厚朴酚处理过的Hela细胞,经Hoechst33258染色后细胞、细胞核出现明显的凋亡形态学变化,在DNA琼脂糖凝胶上可见特征性的梯状条带。结论和厚朴酚具有抑制Hela细胞增殖和诱导凋亡的作用。

Abstract:OBJECTIVE: To assess the effects of honokiol on proliferation and apoptosis of human cervical carcinoma cell line Hela in vitro. METHODS: Cultured HeLa cells were treated with different concentrations of honokiol for the varieties of period (24, 48, 72, 96 h). Cell proliferation was assessed by MTT colorimetric assay. Cell apoptosis was determined by flow cytometry (FCM), Hoechst 33258 fluorescent staining and DNA ladder respectively. RESULTS: MTT assay demonstrated that the proliferation of Hela cells were suppressed significantly by honokiol in dose-and time-dependent manner. FCM analysis showed that the apoptosis rates of Hela cells treated with 10 microg/mL and 20 microg/mL honokiol for 24 h were 22.5% and 62.2%, respectively, while that of the control group cells was 8.7%. After treatment with honokiol, typically morphologic changes of apoptosis were observed by Hoechst 33258 fluorescence staining; Genomic DNA from Hela cells treated with honokiol displayed a characteristic ladder pattern on agarose gel electrophoresis. CONCLUSION: honokiol can inhibit the proliferation and induce apoptosis of human cervical carcinoma cell line Hela.
