不限时相的药物动力学参数估计法

摘    要:本文改变了当前经典的残数法求动力学参数所使用的近似式,把原来只能用消除相数据估计消除速率常数k或β、吸收相数据估计吸收速率常数ka及分布相数据估计分布速率常数α,变成用任何五个等间隔数据就能估计全部参数,所得结果比残数法更准确。本法还取消了残数法要求消除速率常数比吸收或分布速率常数小得多的限制。

Abstract:The classical residual method for estimat-ing pharmacokinetic parameters is some-what complicated: the biexponent process isseparated into two single exponent processesby the approximate re1ation of concentrationwith time and the expression of residualerror. Then the pharmacokinetic paramotersare estimated by twice linear regression.There are too many restriction in samplingtime and size of kinetic rate constants.Sampling has to be made many times in thetime process of absorption, distribution andelimination. The rate constant of eliminationhas to be much less than the rate constantof absorption and distribution. Otherwise, themethod doesn't work. In this article, theauthor presents a new approach to theestimation without incurring the restrictionsmentioned above. Better results are obtainedby this new method because all the mathe-matical expressions used are exact.
