摘    要:目的:研究8-溴-7-甲氧基白杨素(8-bromo-5-hydroxy-7-methoxychrysin,BrMC)诱导人急性T淋巴细胞白血病Jurkat细胞凋亡作用。方法:体外培养Jurkat细胞。FITC标记annexinⅤ/PI染色流式细胞术(FCM)分析细胞凋亡率。ELISA法测定细胞组蛋白/DNA碎片。二氯荧光素二乙酯(2'7'-dichlorofluoresein diacetate,DCFH-DA)荧光探针FCM检测细胞活性氧。结果:BrMC以剂量依赖方式诱导annexinⅤ阳性细胞和组蛋白/DNA碎片增加(P<0.05)。BrMC促进细胞活性氧生成,10 mmol/L N-乙酰半胱氨酸(NAC)预孵育能有效阻断Jurkat细胞活性氧生成,并减弱其诱导细胞凋亡效应。结论:BrMC具有诱导Jurkat细胞凋亡作用,其作用与促进细胞活性氧生成有关。

Abstract:Objective To investigate the effects of 8-bromo-5-hydroxy-7-methoxychrysin(BrMC) on apoptosis of human acute T lymphocyte leukemia Jurkat cell line.Methods Jurkat cells were cultured in vitro.Flow cytometry(FCM) after FITC labeled annexin V staining was used to determine the percentage of apoptotic cells.Histone/DNA fragments were determined using ELISA assay.The generation of reactive oxygen species(ROS) was analyzed by FCM using the fluorescent probe 2' 7'-dichlorofluoresein diacetate(DCFH-DA).Results BrMC significantly induced increase of the percentage of apoptotic cells(P<0.05),and increased level of the histone/DNA fragments in Jurkat cell line(P<0.05),in a concentration-dependent manner.BrMC elevated ROS generation level in Jurkat cells(P<0.05),in a concentration-dependent manner.N-Acetylcysteine(NAC,10 mmol/L) can block generation of ROS and attenuate induction of apoptosis by BrMC in Jurkat cells(P <0.05).Conclusion BrMC can effectively induce apoptosis of Jurkat cells,and this effect is associated with generation of ROS.
