摘    要:TGF-β1/Smad信号传导通路是肾间质纤维化发生发展重要的通路之一。随着分子生物学、中药药理学等研究的不断深入,无论中药单方及其有效成份或复方中药,通过调控TGF-β/Smad信号通路拮抗肾间质纤维化已显现出广泛的药理作用:如直接抑制TGF-β1的产生,或抑制TGF-β1刺激的下游因子CTGF等,或抑制Smads磷酸化,或调节TβR、Smad的表达等,显示出多途径、多层次、多靶点对TGF-β1/Smads通路及信号蛋白的调控效应。但由于复方中药成分复杂,应更深入全面地阐明其对TGF-β/Smad信号通路的各个环节、不同靶点及与其他信号通路相互作用的调控机制,并通过拆方研究、有效部位筛选、有效组分分析等,明确各药物之间的协调作用,以筛选出临床疗效可靠、无不良反应的基础方药及其药物最佳配伍剂量或形式,此为复方中药研究应寻找的最佳突破点之一。

Abstract:TGF-beta1 /Smad signaling pathway is one of the important pathways in development of renal interstitial fibrosis. As the research of molecular biology and Chinese medicine pharmacology,traditional Chinese medicine has displayed a wide range of pharmacological effects in regulating the TGF-beta / Smad signaling pathways antagonism renal interstitial fibrosis. Regardless of traditional Chinese medicine( TCM) monotherapy or traditional Chinese medicine compound and its effective ingredients,performance for: direct inhibition of TGF-beta1,or inhibition TGF-beta1 stimulating downstream factor CTGF,or inhibitory Smads phosphorylation,or regulating the expression of TβR and Smad,which show TGF-beta1 / Smads pathway and the regulation effect of the signaling proteins through multichannel,multi-level and multiple targets. But due to the complex traditional Chinese medicine compound composition,should be more fully discuss the TGF-beta / Smad signaling pathways of each link,different targets and regulatory mechanism of interaction with other signaling pathways,and through research,effective parts selection,effective component analysis and so on,clarify the coordinating role between drugs further,to filter out the basis of clinical curative effect is reliable non-toxic side effects and its clinical drug best compatibility dose or form,this is one of the best breakthrough point of a traditional Chinese medicine compound study.
