中医药调控TGF-β1/Smad信号通路干预肾间质纤维化机制

TGF-β1/Smad信号传导通路是肾间质纤维化发生发展重要的通路之一。随着分子生物学、中药药理学等研究的不断深入,无论中药单方及其有效成份或复方中药,通过调控TGF-β/Smad信号通路拮抗肾间质纤维化已显现出广泛的药理作用：如直接抑制TGF-β1的产生,或抑制TGF-β1刺激的下游因子CTGF等,或抑制Smads磷酸化,或调节TβR、Smad的表达等,显示出多途径、多层次、多靶点对TGF-β1/Smads通路及信号蛋白的调控效应。但由于复方中药成分复杂,应更深入全面地阐明其对TGF-β/Smad信号通路的各个环节、不同靶点及与其他信号通路相互作用的调控机制,并通过拆方研究、有效部位筛选、有效组分分析等,明确各药物之间的协调作用,以筛选出临床疗效可靠、无不良反应的基础方药及其药物最佳配伍剂量或形式,此为复方中药研究应寻找的最佳突破点之一。
