豆腐果苷对大鼠坐骨神经压迫性模型机械痛敏和热痛敏的影响

摘    要:目的:观察豆腐果苷(hel i ci d)对坐骨神经慢性压迫性模型(CCI)大鼠机械性缩足反射阈值(MWT)和热缩足潜伏期(TWL)的影响;探讨豆腐果苷在神经病理疼痛中的调节作用。方法:将32只180~220 g Wi st ar雄性大鼠随机分为4组(n=8);分别为Sham组(假手术组)、CCI组(CCI模型组)、豆腐果苷低剂量组(H1)和豆腐果苷高剂量组(H2)。4组分别于CCI手术前1天、CCI手术后第10天灌胃给药,1次/d,连续10天测定后肢MWT和TWL。结果:CCI手术后第9天Sham组、CCI组、H1组、H2组MWT和TWL值分别与术前基础值比较,Sham组无差异(P〉0.05),CCI组、H1组、H2组均有差异(P〈0.05),CCI模型制作成功。给药后第1~10天,H1组、H2组MWT和TWL值分别与CCI组组间比较,MWT和TWL明显升高(P〈0.05),H2组较H1组保持较高水平。结论:口服豆腐果苷能剂量依赖性提高CCI大鼠机械性缩足反射阈值,延长热缩足潜伏期;可延缓大鼠神经病理性疼痛的形成。

Abstract:Objective: To observe the effect ofhelicid on mechanical withdrawal threshold (MWT) and thermal withdrawal latency (TWL) in rat model with chronic compression injury of sciatic nerve as well as to explore its regula- tion during the development of neuropathic pain. Method: Thirty-two rats were randomly divided into four groups (n=8): sham group (sham operation), CCI group (group of CCI models), low dose group of helicid (HI) and high dose group of helicid (H2). Four groups were administered with drugs intragastrically at the day before operation and the tenth day aider operation, once each day. MWT and TWL of hind legs were tested for ten consecutive days. Result: At the ninth day after CCI surgery sham group,CCI group, Hlgroup and H2 group were compared with basic indexes of MWT and TWL before operation, sham group showed no difference (P〉0.05), CCI group, Hlgroup and H2 group demonstrated differences (P〈0.05), models with CCI were established successfully at the ninth day after CCI opera- tion. During the first day to tenth day after medication, H1 group and H2 group were compared with CCI group respec- tively, indicating that MWT and TWL increased significantly (P〈0.05) as well as H2 group was higher than H1 group. Conclusion: Oral helicid can raise MWT of CCI rat dose-dependently, delay TWL and postpone the development of neuropathic pain of rat.
