淫羊藿总黄酮对去势大鼠股骨骨组织形态学及生物力学的影响

摘    要:目的:探讨淫羊藿总黄酮(TFE)对去势大鼠股骨骨组织形态及生物力学的影响及其机制。方法:采用卵巢切除的方法建立大鼠骨质疏松模型,将60只大鼠随机分为正常对照组,模型对照组,TFE低、中、高剂量组,β-雌二醇组6组,每组10只,分别予相应药物灌胃90天。脱钙完成后将股骨近端沿矢状面切开,制成常规石蜡,切片,光镜下观察骨小梁结构;采用三点弯曲实验机,支点跨距18 mm,加载速度2 mm/min,分别从载荷-变形曲线和应力-变形曲线上获得结构力学参数和材料力学参数。结果:与正常对照组相比,模型对照组大鼠骨小梁出现断裂、间隙增宽、数量减少;与模型对照组相比,各给药组骨组织形态明显改善,以TFE高剂量组最为明显;与正常对照组相比,模型对照组股骨结构力学和材料力学均显著下降,其差异具有统计学意义(P<0.05);与模型对照组相比,TFE低、中、高剂量组及β-雌二醇组生物力学各项指标均上升或显著上升,差异具有统计学意义(P<0.05)。结论:TFE可改善去势大鼠股骨骨组织结构,从而改善其生物力学性能。

Effects of Total Flavonoids of Epimedium on Biodynamics and Bone Morphological Metrology of Ovariectomized Rats

Abstract:Objective: To explore the effects of total flavonoids of epimudium (TFE) on biodynamics and bone morphological metrology of ovariectomized rats and its mechanism. Methods: Rat model with osteoporosis was es-tablished by the excision of ovaries, 60 rats were randomly divided to six groups: normal control group, model con-trol group, low, moderate and high dosage of TFE groups andβ-estradiol group, ten rats each group, all the rats re-ceived intragastric administration of corresponding drugs respectively. After decalcification, proximal femur was cut along vertical plane, and sliced after dealt with traditional paraffin section preparation methods, bone trabecula was observed with light microscope; air bending experimental machine was adopted, pivot spacing: 18 mm, loading rate: 2mm/min, the indexes of structural mechanics and mechanics of materials were obtained by loading-deformation curve and stress-deformation curve respectively. Results: Compared with normal control group, bone trabecula of rats in the model control group showed fracture, gaps broadened and the numbers were reduced; compared with the model control group, the shapes of bone tissues in different medicated group were improved notably, high dosage of TFE group was the most obvious one; compared with normal control group, femur structural mechanics and the me-chanics of materials in the model control group were decreased significantly, the comparison showed significant dif-ference(P〈0.05); compared with the model control group, biomechanical indexes in low, moderate and high dosage of TFE groups andβ-estradiol group were raised or significantly raised, the comparison showed significant differ-ence (P〈0.05). Conclusion: TFE could improve the structure of femur bone tissue in the ovariectomized rats, conse-quently to improve biomechanical capability.
