黄芪多糖对兔动脉粥样硬化内皮细胞功能的影响

摘    要:目的探讨黄芪对新西兰兔动脉粥样硬化内皮细胞功能的影响。方法将40只雄性新西兰兔随机分为4组,每组10只,空白组:正常颗粒饲料;其余3组从实验第1天起给予高脂饲料,牛血清1mL/kg从耳缘静脉注射1次,黄芪多糖(APS)组:每天同时腹腔注射APS(500mg/kg);卡托普利组:同时灌喂卡托普利(5mg/kg),相当于临床剂量的5倍;空白组、模型对照组给予等体积的生理盐水4mL/kg,10周后,测定各组空腹血清或血浆三酰甘油(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C)、一氧化氮(NO)、内皮素-1(ET-1)、丙二醛(MDA)、C反应蛋白(CRP)含量以及超氧化物歧化酶(SOD)活性,并计算主动脉内膜粥样斑块面积。结果模型对照组与空白组比较,TG、TC、MDA、ET-1、NO、CRP明显升高(P〈0.05或P〈0.01),HDL-C/TC比值、SOD活性明显下降(P〈0.05或P〈0.01),主动脉内膜粥样斑块面积较大(P〈0.05);卡托普利组与模型组比较,各项指标无明显差异;而APS组与模型组比较,TG、CRP、NO、MDA、ET-1明显下降(P〈0.05或P〈0.01),TC无明显变化,HDL-C/TC比值、SOD活性明显升高(P〈0.01),主动脉内膜粥样斑块面积明显减少(P〈0.01)。结论黄芪多糖对动脉粥样硬化有明显的预防和治疗作用,可能与抗氧化、免疫调节、保护血管内皮细胞有关。

The Effect of Astragalus Polysaccharide on the Function of Endothelial Cells of in Rabbits with Atherosclerosis

Abstract:Objective To investigate the effect of astragalus polysaccharide(APS) on the function of vascular endothelial cells in rabbits with atherosclerosis.Methods Forty healthy New Zealand white rabbits were randomly divided into four groups.Rabbits in blank group was raised normal diet while rabbits in other three groups were given hyperlipid diet,in additiona with vinous injection of bovine sereum by 1 mL/kg once.Rabbits in astragalus polysaccharide group received intraperitoneal injection of polysaccharide of 500 mg/kg once a day,which replaced by 5 mg/kg captopril in captopril group that equals to 5 times clinical dosage,while rabbits in blank group and model control group were given the same volume physical saline of 4 mL/kg for tollaly 10 weeks.After ten weeks of experiment,the concentration of triglyceride(TG),total cholesterol(TC),high density lipoprotein cholesterol(HDL-C),C-reactive protein(CRP),nitric oxide(NO),maleic dialdehyde(MDA) and endothelin-1(ET-1) were measured.Results Compared with the control group,the levels of TG,TC,MDA,ET-1,NO and CRP were increased significantly in model group(P〈0.05 or P〈0.01),while the activity of SOD and the ratio of HDL-C/TC were remarkably decreased(P〈0.05 or P〈0.01).Compared with model group,the levels of TG,TC,MDA,ET-1,NO and CRP did not changed in captopril group.Compared with the model group,the levels of TG,NO,MDA,CRP,ET-1 and the size of aortic intima atheromatous plaque were significantly decreased in APS group(P〈0.05 or P〈0.01),while the activity of SOD and the ratio of HDL-C/TC remarkably were increased in APS group(P〈0.01).Conclusion APS could effectively accentuate the activity of SOD against oxidation,regulate immunity and protect endothelial function against atherogenesis.APS was a potential drug in treating and preventing atherogenesis.
