三七总皂苷对肺纤维化小鼠肺组织胶原影响的实验研究

摘    要:目的:研究三七总皂苷对小鼠肺纤维化肺组织胶原的影响。方法:C57BL/6雄性小鼠180只,分为假手术组、模型组、三七总皂苷高、中、低剂量组、强的松组,每组30只。除假手术组外其余各组小鼠气管内一次性滴注盐酸博莱霉素,假手术组小鼠气管内一次性滴注等体积的生理盐水。造模后第2天开始给药,持续到处死动物的前一天。三七总皂苷高、中、低剂量组分别为120、60、30mg.kg-1.d-1,强的松组为0.56mg.kg-1,假手术组、模型组分别灌服等体积的生理盐水。各组动物于7d、14d、28d随机处死10只,取肺组织进行HE,Mallory染色,并测定肺组织中羟脯氨酸(HYP)含量。结果:三七总皂苷可降低肺纤维化小鼠肺组织中胶原的含量,减轻肺组织纤维性增生。结论:三七总皂苷可通过降低肺纤维化小鼠肺组织胶原含量而发挥治疗肺纤维化作用。

Abstract:Objective: To study the PNS on pulmonary fibrosis in mice lung tissue collagen.Methods: C57BL/6 male mice were 180,divided into sham operation group,model group,PNS high,medium and low dose group,prednisone group,n = 30.In addition to the sham operation group,the remaining mice a single intratracheal instillation of bleomycin hydrochloride,sham operation group were a single intratracheal instillation with saline.2 days after modeling,starting dose,until the day before the animals were killed.PNS high,medium and low dose group were 120,60,30mg/kg/d,prednisone group was 0.56mg/kg,sham operation group,model group were given equal volume of saline.All the animals at 7,14,28 d were killed 10 lungs were taken for HE,Mallory staining,and determination of lung hydroxyproline(HYP) content.Results: PNS could reduce pulmonary fibrosis in mice with collagen content of lung tissue,reduce the proliferation of fibrous lung tissue.Conclusion: PNS can reduce pulmonary fibrosis in mice with collagen content of lung tissue play a role in treating pulmonary fibrosis.
