中药复方861治疗大鼠肺纤维化的实验研究

为寻找疗效满意、副作用小的中药治疗肺间质纤维化,本实验观察了气管内注入博莱霉素制成的大鼠肺纤维化模型及复方861治疗该模型后第7天和第28天两个时间段的血、肺泡灌洗液(BALF)中透明质酸(HA)和层粘连蛋白(LN)含量变化、病理变化及28天血气变化.观察中药复方861对大鼠肺纤维化的疗效.结果表明:中药复方861可减轻肺纤维化大鼠肺组织的早期肺泡炎和后期肺纤维化(病理);中药复方861可降低模型大鼠早期(7天)血清和BALF中的HA含量及血清LN含量,并可改善动脉血氧分压.因此,中药复方861对大鼠实验性肺纤维化有一定治疗作用.

The therapeutic effect of herbal compound 861 on bleomycin - induced pulmonary fibrosis in rats

The aim of the present study is to test the effect of antifibrosis herbal compound 861 which has been proven effective for liver fibrosis on the experimental pulmonary fibrosis.Methods:male wistar rats were divided into 3 groups.(1)Normal control group:normal saline was given intratracheally and orally.(2)Model group:Pulmonary fibrosis was induced by intratracheal instillation of single dose of bleomycin (BLM) at 5mg/kg.and saline was given orally.(3)861-Treatment group:After intratracheal instillation of BLM,861 was given by oral gavages at a daily dose of 100mg/kg.Rats of all three groups were sacrificed on the 7th,28 th day,respectively,and the lungs were harvested for histopathological examination.The quantitative analysis of histologic changes was performed by gray scale computation scan.The levels of HA and LN in serum in and BALF were determined by radio immunoassay.Artery oxygen partial pressure (P aO 2) was measured on the 28 th day.Results:these results indicated that herbal compound 861 reduce the alvevlitis in the early stage and the fibrosis in the late stage,increases P aC 2,decrease the HA level in serum adn BALF on the 7 th day and the LN level of vein on the 7 th day.It is concluded that herbal compound 861 has therapeutic effecct on bleomycin-induced pulmonary fibrosis in rats.
