针刺对抑郁大鼠行为学及神经内分泌的影响

目的 探讨针刺治疗抑郁症的的作用机制.方法 将48只成年雌雄各半的SD大鼠随机分为正常组、模型组、针刺组和药物组,每组12只.后3组采用孤养和长期不可预见的中等强度刺激应激造成抑郁大鼠模型,同时针刺组和药物组开始治疗,周期21 d.每周测定各组大鼠行为学的改变,治疗结束后用放射免疫法测定大鼠血清ACTH,T_3,T_4,rT_3和TSH的浓度.结果 与正常组相比,模型大鼠行为学评分降低,血清T_3,T_4,rT_3、TSH均有明显减低(P<0.05,P<0.01),而ACTH显著升高(P<0.01);与模型组相比,各治疗组的行为学评分升高,针刺组血清T_3,rT_3,TSH水平均有显著升高(P<0.05,P<0.01),ACTH水平有显著降低(P<0.01).结论 针刺干预可改变抑郁大鼠行为学异常,具有调节抑郁大鼠下丘脑-垂体-肾上腺皮质轴和下丘脑-垂体-甲状腺轴功能的作用.

Effects of Acupuncture on Behavior Change and Endocrine Function in Depressed Rats

Objective To study the therapeutic effects of acupuncture on depression and to probe the mechanism. Methods Forty-eight adult SD rats, 16 females and 16 males, were randomly divided into a normal group, a model group, an acupuncture group and west medicine group, 12 rats in each group. Separated feeding. Long-term unpredictable and middle stimulation stress were used for development of depressed rat model. At the same time, the treatment groups were treated for 21days. The changes of behavior were recorded in every week. The contents of adrenocorticotrophic hormone (ACTH), TSH, T_3, T_4 and rT_3 in the serum were detected by radio-immunity. Results Compared with the normal group,the times of crossing and rearing movement in model group decreased significantly from the 7th day after modeling, the contents of TSH, T_3, T_4 and rT_3 decreased significantly, as well as the contents of ACTH increased significantly in model group. While in comparison with model group, the times of crossing and rearing movement increased significantly in acupuncture and west medicine groups;the contents of TSH, T_3 and rT_3 increased and ACTH decreased significantly in acupuncture and west medicine groups. Conclusion Aupuncture is effective on the depressed rats. It plays the therapeutic role through ameliorate the function of hypothalamus-pituitary-adrenal axis and hypothalamus-pituitary-thyroid axis.
