皱皮木瓜总黄酮对胃肠平滑肌的松弛作用及其机制分析

摘    要:目的探讨皱皮木瓜总黄酮(FLC)对胃肠平滑肌的松弛作用及其相关机制。方法采用家兔离体空肠、回肠和胃底平滑肌生物测定法,观察FLC对空肠自主性收缩和Ach诱导收缩的影响,以及回肠和胃底对不同激动剂(Ach和CaCl_2)的反应。结果FLC对空肠自主性收缩和Ach诱导收缩反应均呈抑制作用;剂量依赖性抑制Ca~(2 )诱导回肠收缩(P<0.01)及Ach所致胃底肌条的收缩,非竞争性拮抗Ach和CaCl_2累积量效曲线,压低最大反应。结论FLC松弛胃肠平滑肌与阻断VDC减少外钙内流和内钙释放作用有关,但与M胆碱受体阻断无关。

Abstract:Objective To elucidate the relaxant effects and related mechanisms of total flavone from Chaenorneles lagenaria Koidz (FLC)on relaxant gastrointestinal smooth muscles.Methods The relaxant effects and related mechanism of FLC were studied on the contraction of isolated rabbit jejunum,ileum and gastric fundus agonized by Ach and CaCl_2.Results FLC inhibited the con- traction of isolated jejunal autonomic and jejunum agonized by Ach.In the isolated rabbit gastric fundus or ileum,FLC were found to inhibit the contraction induced by Ach and CaCl_2 in dose-dependent manner,and non-competitively antagonize the Ach or CaCl_2 cumulative dose-response curves with a lower maximal response,while Atr(atropine)competitively inhibited the gastric fundus contraction induced by Ach.Conclusion The mechanisms of relaxant gastrointestinal smooth muscle of FLC is related to its blocking effect on the voltage dependent Ca~(2 )channel(VDC),but is not related to muscarinic receptor blocking.
