大孔树脂对葛根总黄酮的吸附及分离纯化研究

摘    要:以三峡库区丰富的葛根为材料,研究葛根黄酮的分离纯化工艺。方法利用柱层析技术,比较了4种大孔吸附树脂分离纯化葛根总黄酮的效果。结果用AB-8树脂柱进行吸附洗脱,洗脱液为60%乙醇,流速2.4 ml/min,得到的葛根总黄酮(以葛根素计)纯度为53.5%,AB-8树脂的吸附容量为103.2 mg/g。结论建立了AB-8大孔吸附树脂分离纯化葛根总黄酮的适宜条件,发现AB-8大孔吸附树脂在葛根总黄酮的分离纯化中有好的表现。

Abstract:Objective Flavonoids were purified from Radix Puerarise of Yichang.Methods Four excellent macroretiaclar resins were selected for separating flavonoid in Radix Puerarise.Results The extract was absorbed by AB-8 macroporous absorption resin and was eluted with 60% ethanol at 2.4 ml/min eluting speed.The purity of total flavonoids was 53.5%.The absorptive capacity of AB-8 resin was 103.2% mg/g.Conclusion A feasible technnology was AB-8 for sublimating the total flavonoids in Radix puerariae.
