玉竹多糖脱蛋白方法的比较研究

摘    要:目的 研究玉竹多糖脱蛋白最佳方法.方法 以sevag法、三氯乙酸法、sevag新法、酶法对玉竹多糖分别进行脱蛋白处理,并将两种最佳方法结合对玉竹多糖进行脱蛋白处理.结果 以回归方程计算多糖和蛋白含量,得出玉竹多糖脱蛋白最佳工艺为:玉竹多糖溶液稀释20倍,取50 ml,10%盐酸调pH值至6.5,加入3%体积的胰蛋白酶,65℃保温6h,沸水浴5 min,4 000 r/min离心,去变性酶沉淀,再加入其体积1/5的氯仿和1/25的正丁醇,剧烈振摇20 min,离心,取水相重复操作3次,得脱蛋白液.结论 酶法与Sevag法结合,可降低多糖损失,提高蛋白质脱除率,是玉竹多糖脱蛋白的一种良好的方法.

Abstract:Objective To evaluate the optimal technologies of deproteinization from polysaccharide of Polygonatum odoratum. Methods Based on the contents of polysaccharide and protein,sevag,trichloroacetic acid,sevag new and enzymatic technology were respectively employed.The better two were combined for the further tests.
