摘    要:目的优化金铁锁毛状根液体培养基、碳源、激素和培养条件,促进毛状根生长及皂苷的积累。方法将毛状根置于添加了碳源和植物生长调节剂的液体培养基中培养,观察其形态、色泽变化,测定其生物量和皂苷含量。结果金铁锁毛状根在含有30 g.L-1蔗糖的12MS中生长最快;6-BA、2,4-D均能促进皂苷的合成,但对毛状根的生长有抑制作用;添加少量的NAA、IAA、IBA都能促进毛状根的生长和皂苷产量的提高,当分别添加0.5 mg.L-1NAA、IAA、IBA时,皂苷产量较对照品提高53.42%,25.55%,79.60%。结论通过液体培养基、碳源、激素等因素的改变,可以提高金铁锁毛状根的生长速度和皂苷的含量。

Abstract:ObjectiveTo improve the growth and accumulation of total saponins of Psammosilene tunicoides hairy root in liquid media by optimizing the basal media,carbon sources,plant growth regulators and culture conditions. MethodsTo check the changes of hairy root morphology and colour,and analyze the alterations in biomass and saponin content led by application of carbon source and phytohormone in the liquid medium. ResultsThe formula of 1/2MS was proven to be the best for the hairy root growth,and the addition of 30 g·L-1 sucrose in the media could increase both the biomass and saponin accumulation.Although 6-BA and 2,4-D could improve the accumulation of saponin,both growth regulators exhibited inhibition on the growth of hairy root.In addition,low levels of NAA,IAA,IBA was introduced to the media,respectively,both the growth and accumulation of saponin of hairy roots could be improved to some extent,and addition of 0.5 mg·L-1 of above three regulators,the yield of saponin was increased by 53.42%,25.55%,79.60%,respectively. ConclusionIt can be realized for good biomass and accumulation of saponins of Psammosilene tunicoides hairy roots through optimizing the basal media,and carbon sources and phytohormones.
