摘    要:目的探讨冬凌草甲素对人鼻咽癌细胞CNE-2的抑制作用,为其临床应用提供理论依据。方法SRB法检测冬凌草甲素对CNE-2细胞生长的抑制作用,流式细胞仪分析细胞周期的变化、检测细胞凋亡的发生。结果冬凌草甲素抑制CNE-2细胞的生长,引起细胞死亡;冬凌草甲素诱导CNE-2细胞发生凋亡和坏死,流式细胞分析发现“亚G1期峰”;冬凌草甲素作用后CNE-2细胞的G2/M期比例增多。结论凋亡和坏死同为冬凌草甲素抑制CNE-2细胞生长的机制,可能与改变细胞周期有关,且具有浓度依赖性。

Abstract:Objective To find more effectively chemotherapeutic agents and treatment regimens,we studied inhibiting effects of Oridonin on human nasopharyngeal cancer line CNE-2.Methods Cell growth inhibition mediated by Oridonin on CNE-2 cells was investigated by SRB assay.The effect of Oridonin on apoptotic induction in CNE-2 cells was determined by flow cytometry.Results The growth of human nasopharyngeal cancer line CNE-2 was significantly inhibited by Oridonin in vitro,and the inhibition rates increased by increasing Oridonin concentration,and death cells were mainly necrotic cells and apoptotic cells."Sub-G1" phase peak was found using flow cytometry.Oridonin could cause CNE 2 cells cycle to change.With the increase of drug concentration,G2/M phase cell percentage increased.Conclusion Apoptosis and necrosis are the key mechanism for Oridonin's killing action in human nasopharyngeal cancer line CNE 2,which is partly due to change of cells cycle,and appear to be dose-dependent.
