摘    要:目的:研究黄连解毒汤对脑缺血动物的促智作用并初步探讨其作用机制。方法:采用结扎小鼠双侧颈总动脉合并低血压法制造小鼠脑缺血模型,考察了黄连解毒汤对脑缺血小鼠学习记忆行为(跳台实验、避暗实验)的影响;测定了小鼠脑中丙二醛(MDA)的含量;进一步地测定了大鼠大脑皮质和海马组织中脑血流量。结果:在跳台、避暗实验中,黄连解毒汤能改善脑缺血小鼠学习记忆行为,能显著减少脑缺血小鼠脑中MDA的含量。黄连解毒汤股静脉给药能增加大鼠大脑皮质和海马组织中脑血流量。结论:黄连解毒汤能促进脑缺血动物学习记忆行为,其作用机制与减少脑中自由基终产物MDA含量、增加大脑皮质和海马组织脑血流量有关。

Abstract:Objective:To study improving effects of Huanglian-Jiedu-Tang (HLJDT) on learning and memory in animals subjected to cerebral ischemia and its mechanisms.Methods:Mice were subjected to bilateral common carotid artery occlusion and hypotension. Some experiments were studied including step-down test, step-through test and determination of brain malondialdehyde (MDA) in cerebral ischemia mice. Furthermore, the cerebral blood flow (CBF) of cortex and hippocampus in rat was determined.Results:In step-down test and step-through test, the activity of learning and memory was improved in cerebral ischemia mice by HLJDT. The MDA content of brain in mice subjected to cerebral ischemia was markedly reduced by HLJDT. In addition, the CBF of cortex and hippocampus in rat was significantly increased after rat was administered HLJDT by fermoral intravenous injection.Conclusion:Learning and memory in animals subjected to cerebral ischemia could be improved by HLJDT. Its mechanisms were associated with reduction of brain MDA and increasing the CBF of cortex and hippocampus.
