摘    要:目的 研究五倍子提取物鞣花酸抗乳腺癌MCF-7细胞的生物学活性及机制,探讨乳腺癌防治的新途径.方法 体外培养乳腺癌MCF-7细胞,用五倍子提取物鞣花酸(30,50,70μg/ml)处理细胞48 h后,采用MTT实验分析细胞的增殖;Hoechst33258荧光染料染色法分析细胞的凋亡;流式细胞仪检测细胞周期;Western blotting检测蛋白表达.结果 鞣花酸对MCF-7细胞的增殖有明显抑制作用,随药物浓度增加,抑制率分别为(20.00±4.oo)%、(41.67±2.31)%和( 77.67±0.58)%,与对照组比较有显著意义(P<0.01),呈剂量依赖性;G1期的细胞百分率分别为(54.60±0.67)%、(60.70±3.61)%和(71.90±1.56)%,与对照组(49.60±2.97)%比较,差异有显著性(P<0.01);随药物浓度增加,细胞核致密浓染强蓝色的凋亡细胞明显增多;肿瘤增殖和凋亡相关基因COX-2表达下调.结论 五倍子提取物鞣花酸有抗乳腺癌MCF-7细胞的活性,其机制可能与COX-2下调相关.

Abstract:Objective To investigate the effect of ellagic acid obtained from gall nut on breast carcinoma cell line MCF-7 and its mechanism.Methods Breast carcinoma cell line MCF-7 was treated with 30,50 and 70 μg/ml ellagic acid respectively in vitro.48 h later,cell proliferation was detected by MTT method,cell apoptosis was detected by Hoechst 33258,cell cycle was assayed by flow cytometry,and gene expression was determined by Western blotting.Results Compared with control group,the ellagic acid significantly inhibited the growth of MCF-7 cells in a dose-dependent manner(P<0.01),with the inhibition rate of(20.00±4.00)%,(41.67±2.31)% and(77.67±0.58)%,respectively.Percentage of cells in G1 phase in ellagic acid-treated group was respectively(54.60±0.67)%,(60.70±3.61)% and(71.90±1.56)%,showing significant difference from control group(49.60±2.97)%(P<0.01).The apoptotic cells were increased with ellagic acid levels,but the expression of tumor proliferation and apoptosis related gene COX-2 was decreased.Conclusion The ellagic acid obtained from gall nut has inhibitory effect on breast carcinoma cell line MCF-7,and its mechanism may be associated with declining COX-2 expression.
