柴胡舒肝散含药血清对乳腺癌MCF-7细胞的体外抑制作用及Bcl-2与VEGF的影响

目的探讨柴胡舒肝散含药血清对人乳腺癌MCF-7细胞株体外抑制作用及Bcl-2与VEGF的影响。方法选取SD雌性大鼠48只,随机分为空白对照组、他莫昔芬组和柴胡疏肝散高、低剂量组。利用他莫昔芬片和不同浓度的柴胡疏肝散进行干预,留取含药血清检测MCF-7细胞凋亡,Bcl-2的蛋白表达及血清中血管内皮生长因子(VEGF)的含量。结果柴胡疏肝散组、他莫昔芬组均可以诱导MCF-7细胞的凋亡,其中柴胡舒肝散高剂量组诱导MCF-7细胞凋亡率高于低剂量组和他莫昔芬组,差异有统计学意义(P<0.05);Bcl-2蛋白在干预组中的表达明显低于空白对照组,差异有统计学意义(P<0.05);柴胡疏肝散高、低剂量组及他莫昔芬组、空白对照组中含药血清上清液中VEGF含量两两比较,差异有统计学意义(P<0.05)。结论柴胡疏肝散含药血清可以通过抑制Bcl-2蛋白和VEGF因子的水平来诱导MCF-7细胞的凋亡。

The Inhibitory Effect of the Drug Serum of Chaihu Shugan Powder on the Bcl-2 and VEGF Expression of MCF-7 Cell of Breast Cancer

Objective To study the inhibitory effect of the drug serum of Chaihu Shugan Powder on MCF-7 Cell of breast cancer and the effect of Bcl-2 and VEGF.Methods Forty-eight SD rats were selected and randomly divided into 4 groups with 12 rats in each group.Tamoxifen and the different dose group of Chaihu Shugan powder were used for intervention,and the drug containing serum was reserved for detection.The apoptosis effect on MCF-7 Cell of breast cancer was detected by flow cytometry,the protein expression of Bcl-2 and VEGF were detected by Western Blot and ELISA respectively.Results Both Chaihu Shugan powder group and tamoxifen group could effectively induce apoptosis of McF-7 cells,and the apoptosis rate of MCF-7 cells in the high-dose chaihu Shugan powder group was higher than others,with statistically significant difference(P<0.05).The expression rate of Bcl-2 protein in the intervention group was significantly lower than that in the control group,and the difference was statistically significant(P<0.05).Serum VEGF in the high dose group,the low dose group,the tamoxifen group and the blank control group were significant differences between each two groups(P<0.05).Conclusion The drug serum of Chaihu Shugan powder can induce the apoptosis of MCF-7 cells by inhibiting the levels of Bcl-2 and VEGF.
