摘    要:目的:利用近红外光谱法建立乳块消片生产过程醇提液中总氮的快速定量方法。方法:利用传统的凯氏定氮法测定醇提液中总氮的含量,同时获取其近红外光谱,以模型评价性能参数为指标,优选最佳建模波段和光谱预处理方法,建立总氮含量与NIR光谱数据间的校正模型,对待测样品进行总氮含量预测,实现乳块消生产过程醇提液的快速质量评价。结果:采用PLS方法建立的校正模型,醇提液中总氮含量在0.13~1.94mg·mL^-1范围内,相关系数为0.9707,校正误差均方根(RMSEC)为0.109,预测误差均方根(RMSEP)为0.108。结论:采用信息技术对近红外光谱数据进行处理建立模型,可用于中药制剂生产过程快速质量评价。

Abstract:A method was established to quickly determine the total nitrogen content in Rukuaixiao tablet alcohol extracts using the near-infrared spectroscopy (NIR). The NIR transmittance spectra of the samples were acquired at 4000-10000 cm^-1. Different wave ranges and spectrum preprocessing methods were investigated in line with the evaluation performance parameters. A multivariate calibration model was developed to correlate the spectra, and the corresponding values were determined using Micro-Kjeldahl Method. The study has produced the following results: the correlation coefficient, RMSEC, and RMSEP of the calibration model in the range of 0.13-1.94 mg.mL^-1 was at 0.9707, 0.109, and 0.108, respectively. It is concluded that NIRS, when combined with information technology, makes an effective method for evaluating the quality of Chinese materia medica produced from a manufacturing environment.
