四物汤对血虚模型小鼠补血作用有效部位的筛选

摘    要:目的考察四物汤不同溶剂提取部位对血虚模型小鼠外周血象和免疫器官指数的影响,筛选四物汤补血作用有效部位。方法采用ip环磷酰胺(CTX)复制小鼠骨髓造血功能抑制模型。以驴胶补血颗粒作为阳性对照药物。药物组分别ig给予四物汤的多糖、挥发油、二氯甲烷、乙酸乙酯、正丁醇提取部位。测定外周血象、脾脏指数和胸腺指数。采用HPLC法对四物汤不同提取部位的化学成分差异进行初步分析。结果与对照组比较,模型组小鼠RBC、HCT、HGB、PLT、胸腺指数及脾脏指数明显降低,表明血虚模型复制成功。与模型组比较,四物汤多糖部位可明显升髙RBC、HCT、HGB、PLT、胸腺指数及脾脏指数(P <0.05);二氯甲烷部位可明显升高HCT、HGB、PLT、胸腺指数及脾脏指数(P <0.05);挥发油部位可明显升高HGB、PLT和脾脏指数(P <0.05);正丁醇部位可明显升高HCT、RBC、胸腺指数和脾脏指数(P <0.05);乙酸乙酯部位可明显升高PLT(P <0.05)。四物汤正丁醇部位含有川芎嗪、芍药苷和阿魏酸。二氯甲烷部位含有5-羟甲基糠醛和藁本内酯。乙酸乙酯部位含有5-羟甲基糠醛。挥发油部位含有藁本内酯。结论多糖、二氯甲烷、挥发油和正丁醇部位均为四物汤补血作用有效部位,其中多糖和二氯甲烷部位补血作用最强。

Abstract:Objective To screen the effective parts which can enrich the blood in different solvent extraction of Siwu decoction by investigating the peripheral blood and immune organ index of blood deficiency model mice.Methods The mouse bone marrow hematopoietic inhibition model was replicated with ip cyclophosphamide.The Lüjiao Buxue granule was used as a positive control drug.The model mice in each group were ig given polysaccharide,volatile oil,dichloromethane part,ethyl acetate part and n-butanol part of Siwu decoction.The peripheral blood,spleen index and thymoid index were determined.The diff erence of the chemical compositions in the diff erent solvent extraction of Siwu decoction was analyzed by HPLC.Results To compare with the control group,the RBC,HCT,HGB,PLT,thymoid index and the spleen index have increased,which indicate the blood deficiency model has been replicated successfully.To compare with the model group,the polysaccharide part can increase obviously the RBC,HCT,HGB,PLT,thymoid index and the spleen index(P<0.05).The dichloromethane part can increase obviously the HCT,HGB,PLT,thymoid index and the spleen index(P<0.05).The volatile oil part can increase obviously the HGB,PLT and spleen index(P<0.05).The n-butanol part can increase obviously the RBC,HCT,thymoid index and the spleen index(P<0.05).The ethyl acetate can increase obviously the PLT(P<0.05).The n-butanol part contains tetramethylpyrazine,paeoniflorin and ferulic acid.The dichloromethane part contains 5-HMF and ligustilide.The ethyl acetate part contains 5-HMF.The volatile oil part contains ligustilide.Conclusion The active parts of Siwu decoction for enriching the blood is the polysaccharide,dichloromethane,volatile oil and n-butanol part.And the polysaccharide and the dichloromethane extract are the most eff ective parts.
